Well diffusion test

Mainly used to check if a certain extract has used

Steps

1)Using a sterile cotton swab, P. carotovorum broth culture was swabbed evenly on the surface of a Mueller-Hinton agar plate.

2)Wells were created in the agar and filled with 80 μl of each supernatant.

3)The positive control was bleach and the negative control was 0.85 % saline.

4)Triplicates are performed.

5)The plates were incubated at 30°C overnight and checked for zones of inhibition.

6)The zone of inhibition is measured using a 30 cm ruler in centimetres.
